Résumé : | Lipid exchange between biomembranes takes place in a wide variety of biologicalprocesses such as cell-cell communication or lipid adsorption during digestion. Despiteits importance, the fundamental mechanisms behind lipid exchange are not fully understood. In this Master thesis we will explore lipid membrane-membrane interactions usinglipid model systems such as lipid vesicles and solid-supported planar lipid bilayers. Thekinetics of the interaction between these systems will be monitored by a label free surfacesensitive technique, Quartz Crystal Microbalance with Dissipation monitoring (QCM-D).QCM-D measures real-time changes in adsorbed mass and viscoelastic properties of lipidmembrane layers on solid supports by making use of the inverse piezoelectric effect ofthe support, which consists on quartz coated with metals or semiconductor films. Theeffect of different factors i.e., surface charge, layer composition, vesicle curvature, lipidphase and buffer ionic strength, on lipid exchange will be assessed in terms of kinetic parameters during the exchange process and the phase transition behaviour of the resultinglayers after lipid exchange. Experimental results will be supported by a simple theoretical model which shows the same qualitative figures as the experimental data. This workwill bring insight on the type of mechanism (hemifusion or collision) that drives lipidexchange and its related activation energy |
