Résumé : | The incorporation of adenine-8-14C, guanine-8-14C, uracil-2-14C, orotic acid-6-14C, glycine-2-14C, formate-14C into the acid-soluble fraction, RNA, DNA and proteins of the cells of human uterus carcinoma cells (strain HeLa) has been studied. Results show that extracellular preformed bases are better precursors for intracellular components than extracellular smaller molecules. On the other hand, the percentage of incorporation of intracellular acid-soluble precursors into RNA is higher for guanine than for adenine, and in this respect small molecules appear also to be very efficient precursors. The percentage of incorporation into DNA of acid-soluble formate is much higher than that of any other precursor. HeLa cells thus appear to be able both to utilize directly preformed nucleic acids bases and to synthesize them from smaller precursors. © 1957. |
