Résumé : | 1. 1. Chromatographic investigation of the activation of ribonuclease by potassium cyanide seems to demonstrate that the same active -SH groups act in the different reactions catalysed by this enzyme. 2. 2. On the basis of these results an action mechanism of ribonuclease is proposed; It involves the formation of a PS bond between the substrate and the enzyme. Itshows that the two successive stages of the action of ribonuclease can be considered as essentially similar exchange reactions and also helps to explain the inactivity of ribonuclease towards nucleotides that do not possess a CO group in the α-position of the base-ribose bond. 3. Moreover, it is in accordance with all that is at present known about the RNA-RNase interaction. © 1954. |
