Résumé : | The influence of lithium on psoriasis has been demonstrated by different studies. Lithium can induce a new onset or exacerbate pre-existing psoriasis. Different mechanisms are implicated in the pharmacological mode of action of lithium in relation to the onset of psoriasis. On the molecular level, it is well established that cAMP plays a key role in the pathogenesis of lithium-induced psoriasis. Lithium may also affect the protein kinase C-inositol-biphosphate pathway by an inhibition of inositol-1-biphosphate, resulting in the alteration of cellular transduction signals. On the cellular level, lithium has a mitogen effect and acts by blocking cellular differentiation. It also exerts a chemotactic action on neutrophil polymorphonuclear cells. |
