The design process and the synthetic problems for the construction of conformationally constrained cyclic somatostatins are described. Highly potent analogs containing o-aminomethylphenylacetic acid or o-aminomethylphenylalanine are obtained. © 1995 ESCOM Science Publishers B.V.
