Résumé : The in silico study of protein folding mechanisms and the prediction of the native 3D structure from the sole knowledge of the amino acid sequence have challengend numerous scientists, but have not yet given away all their secrets. In this review, we focus on recent developments in this area, including new views on how folding proceeds and novel methods for bypassing the bottleneck of limityed computer time. We also stress that the ultimate solution to these issues has recently drifted away, when it was realized that some natural proteins may adopt several folded conformations and are not always very robust against mutations.
