Résumé : | The 3 isoenzymes of creatine phosphokinase (CPK) were isolated by chromatography on DE 52 from normal serum and after myocardial injury. After heart infarct the MB form is increased. The Michaelis constants and the optimum pH activity of each isoenzyme were studied. The acidity of reduced glutathione added to the reaction mixture is sufficient to bring the pH value to 6.95 and this increases the sensitivity of the serum CPK determination by about 30%. The determination of the activity at different pH values allows the visualization of the 3 isoenzymes. |
