Résumé : The effect of tri-hydroxymethyl-aminomethane (TRIS) on insulin secretion was studied in vitro by incubation of pieces of pancreatic rat tissue. In control experiments, the rate of insulin secretion progressively increased as the glucose concentration of the incubation medium was raised from 100 to 300 mg. 100 ml. A further increase in insulin output was observed at each glucose concentration by raising the pH from 7.4 to 7.8 by adding NaOH. Added hypertonically at a 25 mM/L concentration, TRIS, did not induce an extra insulin release. The only significant effect corresponded to an inhibition of the insulin release induced by a high glucose concentration. This inhibitory effect of TRIS appeared to be increased in alkaline media and apparently was not due to hypertonicity. © 1970.
